DNA & RNA EXTRACTION KITS

The DNA and RNA extraction kits are categorized under nucleic acid purification (NAP) kits with the objective to improve the speed, ease-of-use, and quality of sample preparation for clinical and high-throughput research applications. The other aim of the kits is to be produced as cheap disposable reagents, with significantly reduced sample volume requirement, which would increase throughput and reduce analysis cycle time. Two kits will be presented 1) ACE Prep Genomic DNA Isolation Kit and 2) ACE Prep Total RNA Isolation kit. The ACE-Prep Kits have been prepared and up-scaled for commercialization. Isolation of DNA and RNA can be performed from blood, body fluid, cultured cells, animal tissue, insects, Gram negative and positive bacteria, yeasts and plants. The methods involved are based on a very simple procedure of selective DNA or RNA precipitation (using detergents and highly effective inhibitors for DNase and RNase) from crude lysates without using hazardous reagents. The precipitation process removes proteins and other cellular components that may inhibit PCR or RT-PCR and other enzymatic reaction but leaves high molecular weight genomic DNA or intact undegraded RNA in solution. The method applied in the ACE-Prep kit uses no spin filters or columns, plus the organic solvents used above. The kits developed are much more versatile in the handling of starting amount of material where a wide range of sample volume can be used, and making it scalable thus gives a chance in handling amounts of samples which maybe an important factor to the isolation. High number of samples can also be prepared simultaneously. In the ACE-Prep Genomic DNA kit, RNase digestion steps are included to destroy RNA. The proteins are precipitated out from the crude lysate. Finally, the genomic DNA is concentrated and desalted by isopropanol precipitation. For the ACE-Prep Total RNA kit, a DNA-Protein Precipitation solution is introduced to precipitate the DNA and protein. At the final step RNA is concentrated and desalted by isopropanol precipitation.
